Sperm live with dependence on the extracellular environment and environmental conditions. Their membranes have proteins and polyunsaturated acids that are easily oxidized by the ROS species that are formed in the cryopreservation process and sperm suffer damage in different cell elements and parameters. Therefore, it is necessary to add antioxidants when freezing sperm. Melatonin and glutation were used as antioxidant agents to reduce oxidative stress. A control group and two experimental groups were established, with and without diluent in the medium. Different parameters were analyzed. Melatonin and glutation were seen to counteract the effect on motility, viability, plasma membrane integrity, ROS species formation and DNA integrity. In addition, adding melatonin with 0.25mM concentration is more effective than 0.1mM. Finally, the importance of the use of diluent in cryopreservation was shown.
